INDICATION

Vrontil 15mg Tab is a multimodal antidepressant indicated for the treatment of major depressive disorder in adults who require intermediate dose titration between the 10mg and 20mg doses.

MECHANISM OF ACTION

At 15mg, vortioxetine achieves higher SERT occupancy and greater serotonin receptor modulation than the 10mg dose, enhancing both serotonergic neurotransmission and the indirect effects on dopaminergic, noradrenergic, and cholinergic pathways. This intermediate dose is used to balance efficacy and tolerability when 10mg is insufficient and 20mg produces tolerability concerns. The procognitive benefits of vortioxetine are dose-dependent and may be more pronounced at 15 to 20mg.

DOSAGE AND ADMINISTRATION

•      Adults: 15mg once daily, with or without food

•      Used as a titration step between 10mg and 20mg, or as a maintenance dose in patients who respond at this level

•      Take at the same time each day

•      Always follow the dosage prescribed by your psychiatrist

SIDE EFFECTS

Common:

•      Nausea (may be more pronounced than at 10mg initially)

•      Headache and dizziness

•      Dry mouth

•      Pruritus

Less common:

•      Sexual dysfunction

•      Insomnia or abnormal dreams

•      Hyponatremia

•      Serotonin syndrome with concurrent serotonergic drugs

NOT RECOMMENDED IN

•      Concurrent MAO inhibitors

•      Known hypersensitivity to vortioxetine

•      Children and adolescents under 18 years

•      Caution with seizure disorders or history of mania

DRUG INTERACTIONS

Same as Vrontil 10mg. MAO inhibitors contraindicated. Rifampicin reduces levels; consider dose increase. CYP2D6 inhibitors increase levels; reduce dose to 5mg. Serotonergic drugs increase serotonin syndrome risk. NSAIDs and anticoagulants increase bleeding risk.
