INDICATION:

 Leflox 500mg is a broad-spectrum fluoroquinolone antibiotic indicated for community-acquired pneumonia, acute exacerbations of chronic bronchitis, acute sinusitis, urinary tract infections (uncomplicated and complicated), acute pyelonephritis, skin and soft tissue infections, and prostatitis caused by susceptible bacteria. It is prescribed to reduce bacterial load, prevent infection progression, and promote rapid clinical recovery. Leflox may be used in hospital or outpatient settings, with therapy tailored according to infection severity and pathogen sensitivity.

MECHANISM OF ACTION:

 Levofloxacin inhibits bacterial DNA gyrase (topoisomerase II) and topoisomerase IV, essential enzymes for DNA replication, transcription, repair, and recombination. Disrupting these enzymes, it causes bacterial DNA strand breaks, leading to rapid bactericidal activity against gram-positive and gram-negative organisms. Its broad-spectrum coverage and potent bactericidal effect make it effective in multiple infections and complex urinary and respiratory tract conditions.

DOSAGE AND ADMINISTRATION:

●     Mild infections: 250mg once or twice daily for 7–14 days, depending on the indication.

●     Complicated UTIs: 250mg once daily for 10 days.

●     Take with or without food; drink plenty of fluids.

●     Complete the full prescribed course even if symptoms resolve.

●     Dose adjustment required in renal impairment.

SIDE EFFECTS:

●     Nausea, diarrhea, headache, dizziness, insomnia, rash, and photosensitivity.

●     Serious: tendon rupture, peripheral neuropathy, QTc prolongation, hepatotoxicity, Clostridium difficile-associated diarrhea.

 Levofloxacin interacts with antacids containing aluminum or magnesium, iron supplements, and sucralfate, reducing its absorption; these should be taken at least 2 hours apart. NSAIDs may increase CNS side effects. Warfarin monitoring is recommended because levofloxacin may enhance the anticoagulant effect of warfarin. Patients should inform their doctor of all concurrent medications to prevent adverse interactions.
