INDICATION

Pasage 20mg is indicated as an adjunct to diet for the treatment of primary hypercholesterolaemia, mixed dyslipidaemia, and hypertriglyceridaemia. It is also used for the primary prevention of cardiovascular events in patients at elevated risk due to age, high-sensitivity CRP, and additional risk factors.

MECHANISM OF ACTION

Pasage contains Rosuvastatin, a statin that works by selectively inhibiting HMG-CoA reductase, the enzyme in the liver responsible for cholesterol synthesis. By blocking this enzyme, the drug reduces the liver's production of LDL (bad) cholesterol and triglycerides, while modestly increasing HDL (good) cholesterol. This helps prevent plaque build-up in arteries and lowers the risk of heart attack and stroke.

DOSAGE AND ADMINISTRATION

•      Adults: 10–20 mg once daily at night, with or without food.

•      Dosage may be adjusted at 2 to 4 week intervals depending on response.

•      Maximum dose: 40 mg once daily.

•      Swallow the tablet whole with water; do not crush or chew.

SIDE EFFECTS

•      Headache, nausea, abdominal pain, or constipation.

•      Muscle pain, weakness, or tenderness (myalgia).

•      Elevated liver enzymes (rare but requires monitoring).

•      Rare but serious: rhabdomyolysis (severe muscle breakdown) or immune-mediated necrotizing myopathy.

NOT RECOMMENDED IN

Contraindicated in patients with active liver disease, unexplained persistent elevated liver enzymes, severe renal impairment, or hypersensitivity to rosuvastatin. Not recommended during pregnancy or breastfeeding. Use with caution in patients with hypothyroidism or a personal or family history of muscle disorders.

DRUG INTERACTIONS

Rosuvastatin may interact with antacids containing aluminium or magnesium (reduce absorption, so take rosuvastatin 2 hours before), cyclosporine, warfarin, fibrates, niacin, and certain antiviral medications. Grapefruit juice interaction is minimal compared to other statins, but caution is still advised.
